Not a difference from the book to the movie (although the movie is very different from the book), but a change between cuts -- Blade Runner was recut to have a happier ending, and a voiceover was added. The DVD restores the director's cut.

The Ice Harvest changes the ending from the book, but most of what precedes that is true to the book.

Robert Altman notoriously changed the endng of The Long Goodbye, but it certainly wasn't to make it more viewer friendly. In fact, it pissed off a whole lot of them.
